Background: Bleomycin hydrolase (BLMH), a homocysteine (Hcy)-thiolactone detoxifying enzyme, is attenuated in Alzheimer’s disease (AD) brains. Blmh loss causes astrogliosis in mice while the loss of histone demethylase Phf8, which controls mTOR signaling, causes neuropathy in mice and humans. Objective: To examine how Blmh gene deletion affects the Phf8/H4K20me1/mTOR/autophagy pathway, amyloid-β (Aβ) accumulation, and cognitive/neuromotor performance in mice. Methods: We generated a new mouse model of AD, the Blmh-/-5xFAD mouse. Behavioral assessments were conducted by cognitive/neuromotor testing. Blmh and Phf8 genes were silenced in mouse neuroblastoma N2a-APPswe cells by RNA interference. mTOR- and autophagy-related proteins, and AβPP were quantified by western blotting and the corresponding mRNAs by RT-qPCR. Aβ was quantified by western blotting (brains) and by confocal microscopy (cells). Results: Behavioral testing showed cognitive/neuromotor deficits in Blmh-/- and Blmh-/-5xFAD mice. Phf8 was transcriptionally downregulated in Blmh-/- and Blmh-/-5xFAD brains. H4K20me1, mTOR, phospho-mTOR, and AβPP were upregulated while autophagy markers Becn1, Atg5, and Atg7 were downregulated in Blmh-/- and Blmh-/-5xFAD brains. Aβ was elevated in Blmh-/-5xFAD brains. These biochemical changes were recapitulated in Blmh-silenced N2a-APPswe cells, which also showed increased H4K20me1-mTOR promoter binding and impaired autophagy flux (Lc3-I, Lc3-II, p62). Phf8-silencing or treatments with Hcy-thiolactone or N-Hcy-protein, metabolites elevated in Blmh-/- mice, induced biochemical changes in N2a-APPswe cells like those induced by the Blmh-silencing. However, Phf8-silencing elevated Aβ without affecting AβPP. Conclusions: Our findings show that Blmh interacts with AβPP and the Phf8/H4K20me1/mTOR/autophagy pathway, and that disruption of those interactions causes Aβ accumulation and cognitive/neuromotor deficits.

Ensiling grass with microbial inoculants is a promising strategy to enhance forage quality, animal performance, and environmental sustainability. This study evaluated the effects of a multi-strain inoculant (Lactobacillus plantarum, L. buchneri, Propionibacterium acidipropionici, and P. thoeni) on silage fermentation, nutrient digestibility, milk production, methane emissions, and rumen microbiota in dairy cows. In a 2 × 2 crossover design, 24 lactating Polish Holstein–Friesians were fed total mixed rations differing only in grass silage treated with or without inoculant. Inoculated silage had lower pH (4.56 vs. 5.06; p = 0.02) and higher crude protein (129 vs. 111 g/kgDM; p < 0.05). Cows fed inoculated silage showed higher ruminal propionate (28.3 vs. 26.3 mM; p = 0.03), reduced ammonia (7.61 vs. 8.67 mM; p = 0.02), and fewer protozoa (1.21 vs. 1.66 × 105/mL; p = 0.03). Nutrient digestibility improved (p < 0.05), while methane emissions declined both per cow (368 vs. 397 g/d; p = 0.01) and per kgDMI (15.1 vs. 16.5; p = 0.01). Milk yield increased (p = 0.04), and the fatty acid profile improved. Our study revealed that cows fed inoculated silage had higher nutrient digestibility, lower methane emissions, and microbial shifts in the rumen detected by 16S rRNA sequencing (p < 0.05).

Nesfatin-1 expression and blood plasma concentration in female dogs suffering from cystic endometrial hyperplasia and pyometra and its possible interaction with phoenixin-14

Abstract Background Nesfatin-1 is a neuropeptide that regulates the hypothalamic-pituitary-gonadal axis and may play a role in uterus function. It is co-expressed with other peptides, such as phoenixin, which can influence sex hormone secretion. Our previous research has confirmed that phoenixin-14 is involved in the development of cystic endometrial hyperplasia (CEH) and pyometra in dogs. Therefore, based on the similarities and interactions between these neuropeptides, we hypothesized that nesfatin-1 might also regulate the reproductive system in dogs. This study aimed to determine the expression of nesfatin-1 and its interaction with phoenixin-14 in dogs with CEH or pyometra compared to healthy females, and concerning animals’ body condition score (BCS 4–5/9 vs. BCS > 5/9). Results The analysis of nesfatin-1 in the uterus of bitches consisted of qPCR, western blot and immunofluorescence assays, and ELISAs. The results showed significantly higher nesfatin-1 encoding gene, nucleobindin-2 mRNA (Nucb2) and nesfatin-1 protein expression in overweight females and those suffering from CEH or pyometra compared to healthy animals. The immunoreactivity of nesfatin-1 was elevated in the uteri of bitches with higher BCS > 5/9. Moreover, nesfatin-1 blood concentrations increased in all examined overweight bitches. In the case of phoenixin signals, we found opposite results, regardless of the female body condition score. Conclusion The etiology of CEH and pyometra are not fully known, although we have expanded the level of knowledge with respect to the possible interaction of nesfatin-1 and phoenixin in female dogs’ uteri. They interact oppositely. With increasing female body weight, the expression of nesfatin-1 in the uterus and its peripheral blood concentration increased. However, for female dogs affected by CEH and pyometra, a decreased level of phoenixin-14, irrespective of their body condition score is characteristic. This knowledge could be crucial in the development of biomarkers for these conditions, which may lead to earlier recognition.

Species and embryo genome origin affect lipid droplets in preimplantation embryos

Mammalian embryo development is affected by multiple metabolism processes, among which energy metabolism seems to be crucial. Therefore the ability and the scale of lipids storage in different preimplantation stages might affect embryos quality. The aim of the present studies was to show a complex characterization of lipid droplets (LD) during subsequent embryo developmental stages. It was performed on two species (bovine and porcine) as well as on embryos with different embryo origin [after in vitro fertilization (IVF) and after parthenogenetic activation (PA)]. Embryos after IVF/PA were collected at precise time points of development at the following stages: zygote, 2-cell, 4-cell, 8/16-cell, morula, early blastocyst, expanded blastocyst. LD were stained with BODIPY 493/503 dye, embryos were visualized under a confocal microscope and images were analyzed with the ImageJ Fiji software. The following parameters were analyzed: lipid content, LD number, LD size and LD area within the total embryo. The most important results show that lipid parameters in the IVF vs. PA bovine embryos differ at the most crucial moments of embryonic development (zygote, 8–16-cell, blastocyst), indicating possible dysregulations of lipid metabolism in PA embryos. When bovine vs. porcine species are compared, we observe higher lipid content around EGA stage and lower lipid content at the blastocyst stage for bovine embryos, which indicates different demand for energy depending on the species. We conclude that lipid droplets parameters significantly differ among developmental stages and between species but also can be affected by the genome origin.

This study investigated the effect of zinc nanoparticles (ZnO-NPs) on the growth parameters, parasitological status, ruminal fermentation, and histopathology of lambs that were experimentally infected with Haemonchus contortus larvae. The infected lambs were divided into two groups ( n = 10/group) and grazed on pasture while being fed one of two diets: a control diet consisting of 350 g/d of a dietary concentrate (CONTROL), and a diet consisting of 350 g/d of concentrate enriched with ZnO-NPs (ZINC). Pasture aboveground plant coverage and plant taxa from phytosociological relevés were used as descriptors to investigate differences in vegetation based on plant medicinal properties and the nutritional value. Communities dominated by plants with medicinal properties were mainly found in the CONTROL group's pasture, while the pasture of the ZINC group contained most plants with outstanding nutritional value. The number of eggs per gram of feces was quantified on days D14, D20, D28, D42, D56, D70, D84, D98, and D107 post-infection. There was a significant decrease in egg shedding from D42 onwards in the ZINC group, and from D56 and D70 onwards in the CONTROL group. The ruminal concentration of ammonia nitrogen ( p = 0.018), n -butyrate ( p = 0.025), n-valerate ( p = 0.002), total protozoa count ( p < 0.001), and the enzymatic activities of α-amylase ( p < 0.001) and xylanase ( p = 0.006) were significantly higher in the ZINC group than in the CONTROL group. The molar proportion of acetate was lower ( p = 0.011) in the ZINC group than in the CONTROL group. Morphological observations of the rumen indicated that the homogeneity of the ruminal papillae was slightly impaired, the lamina propria was inflamed, or lymphocytes had infiltrated. In conclusion, the dynamics of gastrointestinal nematode infection were significantly reduced, probably due to the medicinal and nutritional properties of the pasture plants. This effect was also enhanced by the supplementation with ZnO nanoparticles, which possess strong anthelmintic potential.
